PRIMODIL Tablet

For the first line management of Hypertension, Stable angina, Variant/Prinzmetal’s angina.

PRIMODIL may be used alone or in combination with other antihypertensive and/or antianginal drugs for effective management.

Dosage and administration

Oral: Adults: For hypertension and angina pectoris 5mg once daily. After 2 weeks, dosage may be increased to 10mg once daily.

For patients with hepatic dysfunction, initially, 2.5mg once daily.

In severe aortic stenosis, obstructive cardiomyopathy, Left Ventricular failure.

The common side effects are ankle edema, headache, dizziness, somnolence, fatigue, nausea, flushing and palpitation. Their occurrence is dose-related and found more frequently in elderly. Pruritis, rash, dyspnoea, weakness, dyspepsia, muscle cramps, frequency of micturition/nocturia, cough impotence, asthma, epistaxis, nervousness and conjunctivitis occur less commonly.

Primodil potentiates the effects of thiazides and ACE inhibitors. No interaction with digoxin, cimetidine and warfarin has been reported.

In impaired liver or renal function, CHF, elderly, children and nursing mothers. Amlodipine in combination with beta-blockers is generally well tolerated but should be used cautiously while administering intravenously in patients associated with impaired cardiac left ventricular function.

Amlodipine is a long acting, vaso-selective calcium antagonist structurally related to nifedipine, but with binding and pharmacologic properties that uniquely distinguish it from other agents of this class. It inhibits calcium influx into cardiac and vascular smooth muscle via-L-type calcium channels. It has a sustained gradual onset of antihypertensive effect. It preserves the circadian patterns of blood pressure changes and has favourable effects on cardiac output. It has favourable effects on renal haemodynamics and does not affect insulin levels. It has shown antithrombotic potential.
