PILO Eye drops

It is used as direct acting miotic.

To control IOP in glaucoma. For emergency relief of angle closure glaucoma.

Dosage

Instill 1 drop 2-4 times daily. Patient response may vary.

Hypersensitivity to any component of the formulation.

Transient symptoms of stinging and burning may occur. Conjunctival vascular congestion and true allergy may occur but are unusual. Ciliary spasm, temporal or supraorbital headache, and induced myopia may occur, all consequent to drug-induced contraction of the ciliary muscle. Systemic toxicity following appropriate topical ocular administration is extremely rare.

No reports of its use in pregnancy and lactation have been located. This drug fails under category C.

Caution is advised in patients with acute cardiac failure, bronchial asthma, peptic ulcer, hyperthyroidism, GIspasm, urinary tract obstruction, Parkinson’s disease, recent MI, hypertension or hypotension.

PILO produces pupillary constriction, stimulate the cilliary muscles and increase the aqueous humor outflow facility. Miosis produced through the contraction of the iris sphincter, causes increases tension on the scleral spur (reducing outflow resistance) and opening of the trabecular mesh work spaces facilitating outflow. With the increase in outflow facility there is a decrease in intraocular pressure. PILO penetrates the cornea well and produces a low incidence of allergic reactions. It is inactivated by tissues of the anterior segment of the eye, partly by reversible binding of the drug to tissues but also by appreciable enzymatic hydrolysis to the primary metabolite, pilocarpic acid. The mean elimination half-life is 0.76 hours following 5mg 3 times per day for 2 days.
